摘要(Abstract):
目的 基于网络药理学探究丹参治疗多发性骨髓瘤潜在作用及分子机制。方法 通过中药系统药理学数据库与分析平台(TCMSP)筛选并获取丹参的主要活性成分及其对应的作用靶点,利用 Gencards数据库检索并收集多发性骨髓瘤的主要作用靶点,通过线上分析工具 VENNY 获取丹参与多发性骨髓瘤的共同靶基因。通过 STRING 数据库构建蛋白质-蛋白质相互作用 (PPI)网络,并运用 Cytoscape_v3.10.3软件及其“CytoNCA”插件对网络进行拓扑分析,筛选出核心靶点。在可视化综合发现数据库(DAVID)中对药物-疾病交集靶点基因进行基因本体论(GO)功能注释和京都基因与基因组百科全书(KEGG)通路富集分析。结果 从丹参中得到40种活性成分及609个有效靶点,作用于多发性骨髓瘤的主要活性成分为木犀草素、丹参素B、香紫苏醇等,关键靶点为蛋白激酶B1(AKT1)、非受体酪氨酸激酶基因(SRC)、肿瘤坏死因子(TNF)等,通过调控坏死性凋亡通路、嘌呤代 谢通路、人乳头病毒感染通路等通路发挥作用。结论 丹参通过其活性成分木犀草素、丹参素 B、香紫苏醇等调节坏死性凋亡通 路、嘌呤代谢通路,并通过AKT1、SRC、TNF等核心靶点发挥对多发性骨髓瘤的保护作用。
